Yes, definitely, but different extensions have different impacts, and while some only affect Chrome when you use them (e.g. when you right click and choose them), others take a toll on Chrome all the time. You can use Chrome's task manager (Shift Esc) to get an idea of the impact of the extensions at any particular time.

No, a disabled extension does not run, and therefore does not use computer resources in the same way. It may still have some residual effect because they mentioned in various configuration settings and they add to the file count, but that effect will be negligible.
